Measurement of Sperm Concentration in Stallion Ejaculates Using Photometric or Direct Sperm Enumeration Techniques
نویسندگان
چکیده
A highly significant correlation was detected among photometric and direct sperm enumeration techniques for evaluating sperm concentration. Depending on the instrument used, the photometric techniques tended to overestimate or underestimate sperm concentration when compared to a hemacytometer standard. Precautions should be taken when using techniques other than a hemacytometer to measure sperm concentration, especially for stallions with large mare books where the ejaculate will be split among multiple mares for insemination. Seasonality and freezability vs routine parameters in stallion semen.
The fertilizing ability of stallion semen was analyzed using fresh and frozen samples, obtained before (June-July) or during (October-November) the breeding season. Thirty ejaculates obtained from 4 stallions were used. The analysis comprises routine seminogram; ATP concentration (Comhaire et al., 1983); subjective and objective motility and sperm velocity (Makler, 1980). Freezing was done foll...
متن کاملStallion Sperm Selection by Density Gradient CentrifugationInvolving a Double Layer Colloid: Effects on Sperm Subpopulation Dynamics in Fresh and Stored Semen
Sperm selection techniques have became an important tool to improve sub fertile ejaculates. The main aim of this study was to assess the dynamics of motile sperm subpopulations of ejaculates from subfertile stallions subjected to density gradient centrifugation (DGC) involving a double layer colloid [DGC2], before and after storage for 24 and 48 hours. Ejaculates from eight stallions with ferti...

BACKGROUND Artificial insemination is not as widely used in horses as in other domestic species, such as dairy cattle and pigs, partly because of the wide variation in sperm quality between stallion ejaculates and partly due to decreased fertility following the use of cooled transported spermatozoa. Furthermore, predictive tests for sperm fertilising ability are lacking. The objective of the pr...
متن کاملNew Methods for Selecting Stallion Spermatozoa for Assisted Reproduction
Improved sperm selection techniques are needed to increase the efficiency of equineassisted reproduction. Single layer centrifugation (SLC) of spermatozoa has been shown to improve the quality of stallion sperm samples. In this study, the functionality of selected stallion spermatozoa was tested by intracytoplasmic sperm injection of equine oocytes after selection by SLC through Androcoll-E or ...
متن کاملAn analysis of Selected Aspects of Sperm Quality in Fresh and Cooled-Storage Stallion Semen
This review summarises the most significant findings related to the issue of the quantitative and qualitative indicators of stallion ejaculates, its processing when producing chilled insemination doses and the possibility of influencing the individual steps of the entire process. Artificial insemination, with cooled-storage semen, is increasingly used in horse breeding because it offers many ad...
